Following a successful bid to the government as part of a nationwide expansion of medical student numbers in the UK, the Peninsula Medical School opened its doors on August 1, 2000, six years before the dental school. Professor Sir John Tooke, who at the time held a combined appointment at Peninsula Medical School and the Royal Devon and Exeter Hospital, was the creative force behind the application.

Peninsula Medical School's first dean was Professor Tooke, who held the position until the autumn of 2009. He was appointed chair of the UK Committee of Heads of Medical Schools and was awarded a knighthood in the New Year's Honours list for 2007 in recognition of his vision and drive. As part of an effort by the British government to increase the number of medical professionals in the country, Peninsula Medical School was established. According to the league tables in the media, Peninsula Medical School (PCMD) has not only consistently surpassed the other "new" institutions but has also proven to be extremely competitive versus existing medical schools. This is according to the school's performance.

The Peninsula Medical School started the process of splitting apart in January 2013, which resulted in the formation of the Plymouth University Peninsula Schools of Medicine and Dentistry as well as the University of Exeter Medical School. The teaching of medical students also took place at the Peninsula Medical School in Barnstaple, Today, in general practices across the region, as well as in a number of community-oriented healthcare settings.

Peninsula Medical School welcomed its first intake of 130 undergraduate students on September 30, 2002. Since September 2003, Peninsula Medical School's annual intake has risen to 167. Peninsula Medical School received funding for further expansion in January 2006, resulting in an increase in both UK and overseas placements. In September 2006, 214 students enrolled at Peninsula Medical School; by September 2010, that number had climbed to 230.
